Aneuploidy rates and likelihood of obtaining a usable embryo for transfer among in vitro fertilization cycles using preimplantation genetic testing for monogenic disorders and aneuploidy compared with in vitro fertilization cycles using preimplantation genetic testing for aneuploidy alone

This study sought to compare aneuploidy rates among in vitro fertilization (IVF) cycles using preimplantation genetic testing for monogenic disorders (PGT-M) and aneuploidy (PGT-A) compared with IVF cycles using PGT-A alone, and to determine the likelihood of obtaining at least one usable embryo in cycles using PGT-M+PGT-A compared with cycles using PGT-A alone. In vitro fertilization cycles using PGT-M+PGT-A have similar age-specific aneuploidy rates to those using PGT-A alone. Cycles for patients ≤40 using PGT-M+PGT-A are significantly less likely to yield a usable embryo compared with those using PGT-A alone.

Validation of a mail-in delayed semen analysis protocol developed for home collection

This study sought to validate a mail-in delayed semen analysis service using deidentified remnant samples from a US fertility clinic. In a controlled laboratory setting, total motility, concentration, and morphology measures were highly consistent between the 1- and 26-hour analyses. In a real-world setting, which included commercial shipment of specimens, the mean %CVs for total motility and concentration were 15% and 27%, respectively, which were more variable than those in a controlled laboratory setting, yet still considerably less variable than that measured between laboratories using College of Anatomical Pathologist proficiency testing during the study period. These results demonstrate the validation of a laboratory service that provides accurate, comprehensive semen analysis on specimens collected remotely and shipped overnight to a clinical diagnostic laboratory.

Unraveling the genetic basis of azoospermia: transcriptome profiling analyses in a Greek population

This study investigates whether idiopathic nonobstructive azoospermia (iNOA) has its own transcriptomic signature. A significantly altered transcriptomic profile of protein-coding genes was identified in the testicular tissues from men with iNOA. A total of 3,858 genes exhibited dysregulated expression, with 1,994 genes being exclusively downregulated and 1,734 upregulated. A distinct messenger RNA expression profile and altered transcriptomic activity were identified in the testicular tissues of men with iNOA.

Stressed out: how forces from uterine contractions influence fibroid progression, a Narrative Review

Uterine fibroids are exposed to significant mechanical forces due to routine, monthly uterine contractions and have also been found to generate contractions in the junctional zone or inner myometrium. These are not the only mechanical forces that uterine fibroids experience but also compression and strain, or percent change in length, due to the stiff extracellular matrix of the fibroids. The forces may vary by location within the tumor. Strong uterine contractions not only cause pain but may also contribute to uterine fibroid growth, which, in turn, may lead to worsening symptom severity. This review discusses uterine contractions in the nonpregnant uterus and what is known about the impacts of mechanical forces on uterine fibroid cells.
